Exopolysaccharide production from Alkalibacillus sp. w3: statistical optimization and biological activity

Abstract Microbial exopolysaccharides (EPS) provide a broad range of applications. Thus, there is an increasing interest in the production, characterization, and use EPS derived from various microorganisms. Extremophile polysaccharides have unique properties applications due to its structures. The importance synthesized by new bacterial strain, Alkalibacillus sp. w3, was highlighted this study. haloalkalitolerant firmicute that recovered salt lake, optimized for biological activities were studied. Exopolysaccharide synthesis observed Horikoshi I broth medium. optimal culture conditions achieving highest exopolysaccharide production 7-day incubation period, pH 10, 250 g/L NaCl. most effective carbon nitrogen sources glucose combination yeast extract peptone. Additionally, Plackett-statistical Burman’s design showed all factors tested had favorable impact, with having greatest significance on EPS. model’s best predictions resulted two-fold improvement compared original yield before optimization. contained 65.13% carbohydrates, 30.89% proteins, 3.98% lipids. Moreover, produced w3 demonstrated anticancer activity against hepatocellular carcinoma (HepG2) human colon (HCT-116) cell lines, IC50 values as low 11.8 15.5 µg/mL, respectively, besides antibacterial Gram-positive, Gram-negative bacteria, yeast. Based these results, made has many useful properties, which make it suitable medical field.
